A second season has begun production. It's said to take place 10 years after season 1. This will likely conclude the story.
Actually, John Blackthorne is supposed to be communicating with Mariko in Portuguese. As it's a language they both understand. Until John learns Japanese well enough to not need a translator. We simply hear/see them speaking English.
Unfortunately, no. It's possible it will, as Disney/FX has been releasing some of their more recent shows on bluray (such as Justified: City Primeval). But given the length of time since it aired, it's not looking promising.
