Whoopi Goldberg's daughter, Alex Martin, appears in this movie as one of the classroom children. She says the "Yo Mama" line about Skittles.
When the students are on the basketball court, Rita (Lauryn Hill) starts rapping. Her rapping was improvised.
The famed high note performed by Ryan Toby (Ahmal) during the "Oh, Happy Day" song is a high E-flat, one of the highest and toughest notes to hit for singers and vocalists. Toby was sixteen years old during the shooting of this movie.
Internet rumors persisted that Whoopi Goldberg was very unhappy making Sister Act (1992), and made this sequel only after Disney decided to finance her dream project, Sarafina! (1992). However, "Sarafina!" debuted at Cannes before "Sister Act" opened in theaters.
Thomas Gottschalk (Father Wolfgang), who is a famous German television host, appeared as a consequence of Whoopi Goldberg losing a bet in Thomas' game show Wanna Bet? (1981).

Bill Duke: As Mr. Johnson, Sketch's (Ron Johnson's) boss who admonishes him for spraying graffiti and chatting with his friends, when he should be working.
