PLAP Antibody (8B6) is a mouse monoclonal IgG2a kappa light chain antibody that detects the PLAP protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). PLAP (8B6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The placental alkaline phosphatase (PLAP) protein, also known as ALPP, is a 530 amino acid glycoprotein that plays a crucial role in cellular processes by catalyzing the hydrolysis of phosphomonoesters into inorganic phosphate and alcohol, which is vital for various metabolic pathways. PLAP is primarily expressed in the placenta and is also found in the serum of pregnant women, with ectopic expression observed in certain cancers, such as ovarian and testicular tumors. PLAP expression in these tissues suggests involvement in guiding migratory cells and facilitating the transport of specific molecules, including fatty acids and immunoglobulins, across the plasma membrane, which is essential for maintaining cellular homeostasis and supporting tumor progression. Notably, PLAP, along with germ cell alkaline phosphatase (GCAP) and intestinal alkaline phosphatase (IAP), are tissue-specific alkaline phosphatases that share 90-98% homology and are clustered on chromosome 2q, highlighting their evolutionary relationship and functional significance in human physiology.

SC has two antibodies for human PLAP (G-9 and 8B6). 8B6, is more widely cited, but why is the single band seen by WB not 70kDa (dimer?) while many bands for G-9? Also do you have any blots showing KD with your siRNA to PLAP (sc-38919) for either antibody?

Thank you for your question. There are three forms of the PLAP protein that can be detected with these antibodies (~67 kD=monomeric,~130 kD=dimeric & ~180 kD=trimeric). We generally see the monomeric or dimeric in our sampels. Since PLAP expression differs in tumor cell lines, what form will be seen varies depending on the samples. Unfortunately we do not have siRNA data for this target with these antibodies, so we are unable to provide that.

Can PLAP (8B6): sc-47691 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, PLAP (8B6): sc-47691 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
