ACT 17
1. Describe other tests to detect RF. Discuss their principles
Other test to detect the Rheumatic factor are the following:
⮚ Latex agglutination method.
-It is a rapid latex agglutination test for the qualitative and semi-quantitative determination of
Rheumatoid Factor (anti-gamma globulin)
Principle:- Latex particles coated with human gamma globulins are allowed to react with test serum.
Agglutination will be observed if RF is present in the serum.
⮚ Haemagglutination test
-It is a type of agglutination test performed on red blood cells.
Principle:- It mixes the blood being tested with a sheep’s red blood cells that have been covered with
rabbit antibodies. If RF is present, the red blood cells agglutinate.
⮚ Nephelometry test
-A sensitive technique for measuring the degree of light scatter caused by the interaction of rheumatoid
factors with heat aggregated human IgG.
Principle:- Nephelometric testing of rheumatoid factor levels is based upon antigen-antibody reactions.
RF binds to IgG to form immune complexes. As the number of immune complexes grows, the solution
becomes more visibly “cloudy”.
⮚ Other tests to determine past streptococcal infections have been developed including the anti-
CHO test (possible use for rheumatic fever).
