RESEARCH EXPERIENCE

I. Looking For The Probable Halogenase Involved In Halogenation Of Signal


Molecule.

From July 2007 onwards I worked on "Halogenase Involved in Chlorination of Dif-1


Molecule in Dictyostelium". Advisor: Dr. Rajesh S. Gokhale from National Institute
of Immunology (NII) , JNU Complex, New Delhi-110067.

Brief Project methodology:


· Bioinformatics analysis to find the probable genes in dictyostelium genome on the
basis of their homology to already characterized halogenase by other groups.
· Cloning of these genes in different expression vectors.
· Check the expression of the protein and to get the protein in soluble fraction.
· To purify the protein of the contaminating proteins using FPLC.
· To put up assays with probable substrate and to check the product formation on TLC
and HPLC to confirm.
· Analysis of the mass of the sample with the help of Mass spectrometry (LC-MS) and
MALDI-TOF-TOF.
· Analysis of the results.

II. Looking For A Novel Drug Target Against Mycobacterium tuberculosis Aiming
At Vaccine Development

I was associated with another project which is related to "Evolutionary Studies of the
Gene Cluster in Various Species of Mycobacteria".

Brief project methodology:


· Retrieving the gene sequences from respective database and PCR amplification using
genomic DNA
· Cloning and expression of the PKS genes present in the cluster of
M. paratuberculosis.
· Protein expression studies and purification of the protein.
· Radioactive assay with probable substrate and identification of the product by using
TLC, HPLC and finally mass-spec analysis.
· Analysis of results.

M. Sc. Thesis project

From this project, we have got some interesting findings which are going to publish
soon. One has already been communicated in Journal of Food Biotechnology.

Biotechnological productions of Lactic Acid from potato processing waste water by
Lactobacillus plantarum isolated from Fermented Green Chutney's
International Conference on Biotechnological Engineering ICBioE'07,
Kaula Lampur, Malaysia, 8 - 10 May 2007.

From Jan 11th 2006 to June 30th 2006 for partial fulfillment of the Masters program in
Biotechnology. Six months project work entitled "Value Added Products from W aste
Stream of Industry Effluent''. Advisor: Dr. Abhijit Ganguli, Thapar Institute of
Engineering and Technology, Patiala, Punjab.

Lactic acid bacteria possessing amylases can utilize starch fermentatively for lactic acid
production. Amylolytic lactic acid bacteria (ALAB) account for a substantial portion in
different types of foods and are widespread among the non-dairy food environments and
different geographical areas. Some lactobacillus species have been reported to be able to
reduce the hydroxyl group of the polysaccharides into aldehyde group and finally the
other group of bacteria from lactobacillus genera to convert this aldehyde group into an
acid moiety. For these biochemical conversions usually we would require more then one
microbe and also require setting up a multiple fermenter to carry out the two steps
separately which is quite cost intensive. So the idea was to find an organism which could
carry out both the steps of this conversion of starch from waste effluent into lactic acid.

Brief project methodology:

· Screening of the Lactobacillus sp. bacteria which could utilize starch as energy
source
· Starch degradation profile on nutrient medium
· Growth optimization in different media
· Quantification of the amylase activity by lactobacillus plantarum in nutrient
medium
· Temperature and pH optimization for lactic acid production by Lactobacillus
plantarum
· Starch degradation profile in nutrient medium using a dialysis sac
· Morphological & biochemical characterization of the strain

Summer Training

From June 1st 2005 to July 15th 2005 as a 45 days summer training project work
entitled "Studies On Glutamate Dehydrogenase from Strain PPD
(Pseudomonas sp.) Grown On Phthalate Isomers". Advisor: Dr. P. S. Phale,
School of Biosciences and Bioengineering, Indian Institute of Technology-
Bombay, Mumbai. The work was published as Biodegradation of phthalate
isomers by Pseudomonas aeruginosa PP4, Pseudomonas sp. PPD and
Acinetobacter lwoffii ISP4 in Applied Microbiology and Biotechnology. 2006
Oct; 72(6):1263-9. Epub 2006 Apr 11.

This lab is basically a metabolic engineering lab and the work is mainly focused on
studying biodegradation of phthalates. Our aim was to look at the enzymatic activity of
GDH (Glutamate Dehydrogenase) produced by a strain of Pseudomonas sp. which
actually is responsible for cleavage of closed ring structures and to convert them into
aliphatic compounds so as to enhance the biodegradation of these recalcitrant compounds.
This study included growing this strain on different phthalate isomers and study their
growth profiles and also to see the effect of addition of glutamate on their profiles in
minimal salt media and then looking for the enzyme activity using spectrophotometer and
performing activity staining (a substrate-enzyme based activity reaction on native PAGE).
