To prepare 100 mL of a 1% starch solution, follow this step-by-step procedure:

🌾 Ingredients and Materials:

 Starch powder (soluble starch or corn starch)


 Distilled water
 Beaker (100–250 mL)
 Glass rod for stirring
 Hot plate or water bath
 Measuring cylinder
 Funnel and filter paper (optional)

📏 What does 1% w/v mean?

 1% w/v = 1 gram of starch in 100 mL of solution

🧪 Procedure:

1. Weigh starch:
Measure 1 gram of starch powder.
2. Make a paste (to avoid clumping):
In a small beaker, add 10 mL of cold distilled water to the starch and stir it to form a
smooth slurry or paste.
3. Heat remaining water:
Heat 90 mL of distilled water to near boiling (~90°C) using a hot plate or water bath.
4. Add paste to hot water slowly:
While stirring continuously, add the starch slurry to the hot water. Keep stirring.
5. Continue heating and stirring:
Boil the mixture for 3–5 minutes until the solution turns clear or slightly
translucent, indicating the starch is gelatinized.
6. Cool and adjust volume:
Let the solution cool to room temperature, and transfer it to a 100 mL volumetric
flask or measuring cylinder.
Add distilled water to make the total volume exactly 100 mL.
7. (Optional) Filter:
If any undissolved particles remain, you may filter the solution using filter paper.

 Use the solution fresh, as starch solutions can degrade or become


contaminated over time.
 Store in the refrigerator if not using immediately, and use within
24 hours for enzyme assays.
