Yes, he will. He still teaches you everything you need to know about being a Spider, so no need to worry!
Yes. The only person that isn't voicing her character is Kirsten Dunst (Mary Jane). James Franco WILL be voicing Harry Osborn and J.K Simmons WILL be voicing J. Jonah Jameson contrary to the previous games. Unless if you include Michael Clarke Duncan, who played the Kingpin in the 2002 Daredevil film.
Also, due to her character not appearing in the game, Bryce Dallas Howard does not voice Gwen Stacy.

Yes, it will. In fact, this will be Spider-Man 2 on steroids. The city is larger and much more detailed. You will have freedom beyond your wildest dreams!
Because there are two different versions of the game, there are different villains on certain consoles. They are listed as follows.
Wii/PS2/PSP: Morbius, Shriek, Sandman, Venom, New Goblin, Kraven, Lizard (different design than PS3/360), and Carlyle.
However, Rhino will be available on the PS2 version.
X360/PS3/PC: Sandman, Venom, New Goblin, Kraven, Lizard (different design than Wii/PS2/PSP), Carlyle, Scorpion, Kingpin, Calypso, and Rhino.
Morbius and Shriek are exclusive to Wii/PS2/PSP while Kingpin, Calypso and Scorpion are exclusive to the X360/PS3/PC version. As can be seen above, Lizard will have a different design on each version. It's unknown exactly what the design differences will be.

The answer to that isn't very clear. Basically, what's been said by Activision is that Treyarch developed the X360/PS3 version and Vicarious Visions developed the Wii/PS2/PSP version. The PC version was ported from the 360 by Beenox, so it's identical to that version.
Ok, now this is where it gets kind of tricky. Activision have made it very clear that they want money by not only creating different versions between current and previous generation consoles, but between all of platforms as individuals. What I'm trying to say is that the game is different on EVERY console/platform it was developed for.
All the differences are as follows:
Stuff in every version of the game -
* Large, open ended world, the world is 2.5x the size of SM2 for all platforms (excludes GBA and is a little smaller than 2.5x on the DS).
* Symbiote Suit's Rage System (excludes DS and possibly GBA).
* Movie storyline.
* When the game is 100% completed, there is a secret unlockable (unknown if also for GBA and DS versions).
Wii -
* Unique controls featuring the Nunchuk and Wii Remote.
* Morbius and Shriek as bonus villains (confirmed here)
* Ability to switch between the symbiote suit and traditional suit at will.
* Excessive use of the symbiote suit and high crime rate will effect the city (dark and moody clouds, trash build-up, grumpy citizens, etc). Alternatively, if you fight crime and do good, the city will clean itself up.
* Pressing the C button will re-center the camera behind Spider-Man.
* Holding the C button and moving the Nunchuk horizontally or vertically will allow you to look around freely.
* Cineractives will use motion-controls.
* Cineractives will take place in different places than the 360/PS3 version.
