Quickly Operation Guide
Replace Chromatographic Column
1. Open the chromatographic column oven, unscrew the tight joints at both ends of the
chromatographic column, then take out the old chromatographic column.

2. Install the new chromatographic column, screw the tight joints at both ends (the arrow
direction indicated on the label attached to the chromatographic column).
3. Close the chromatographic column oven.
4. Click “ ” icon in the Test interface -> “Replace” -> Swipe your card ->
Click “Yes”.

5. Setting the temperature which is attached to the chromatographic column (30~40℃).

6. Select one of the two methods to complete whole blood infusion according to the
prompts.

(1) 40μL whole blood added into a 1.5mL EP tube -> Put in the STAT port -> Click “Yes” ->
Shake well and cut off the tube cap -> Click “Yes” -> Perform whole blood infusion.
(2) 40μL whole blood and 600μL hemolytic agent L added into a 1.5mL EP tube -> Shake
well and cut off the tube cap -> Put in the STAT port -> Click “Next” -> Perform whole blood
infusion.
7. Wait about 1.5 minutes, the analyzer will complete the chromatographic column
replacement procedure.
Replace In-line Filter
1. Rotate the handle and pull out the filter bracket.

2. Take off the old filter, put in the new filter.

3. Click “ ” icon in the Test interface -> click “Replace” ->click ”Yes”.
4. Wait about 10 minutes, the analyzer will complete the filter replacement procedure.
Replace Reagent
■ Scanning the reagent QR code
1. Replace a new reagent.
2. With the external barcode reader, scan the QR code on the label of the reagent, then click
“Yes”.

3. Wait about 1.5 minutes, the analyzer will complete the reagent replacement procedure.
4. The same goes for the replacement of eluent B, C, D and also hemolytic agent L.
■ Enter manually
1. Replace a new reagent and click “ ” icon.
2. Click “Replace” -> Input the Lot -> Click “Yes”
3. Wait about 1.5 minutes, the analyzer will complete the reagent replacement procedure.
4. The same goes for the replacement of eluent B, C, D and also hemolytic agent L.
Replace Recording Paper
1. Pull the printer lever outward to open the printer door.
2. Load recording paper (recommended width is 57mm) into the paper cavity in the direction,
with the paper end positioned outside the paper outlet .

Maintain Probe
■ Manually cleaning
1. Click the “Maintain Probe” in the System interface.
2. Click “Yes”.
3. Place a W.B. tube with 5mL cleaning solution in STAT Port.

4. Click “Yes”. Wait about 5.7 minutes, the analyzer will complete the maintain probe
procedure.
■ Automatic cleaning
1. Click the “Settings” button in the System interface -> Click the “Maintain Position” button. In
current mode, the operator can’t perform emergency sample test !
2. Place a W.B. tube with 5mL cleaning solution in STAT Port.

3. The analyzer will execute sampling needle cleaning procedure each round of test.
Calibration
1. Take out the calibrators from 2~8℃ and recover the room temperature for about 10~15
minutes.
2. The Lot and target value can be automatically input by scanning the QR code on the external
box of the calibration with the external barcode reader, or manually input in the Calibration
interface.
3. Add 100uL of hemolytic agent L to dissolve calibration materials, shake well and let stand for
30 minutes.
4. Conduct pre-dilution according to the ratio of 10:1500 (add 1.5 mL hemolytic agent L to 10uL

calibration materials). Shake well and cut off the tube cap.
4. Put the EP tube with diluted calibration materials into the 1st and 2nd position of the
calibration rack (marked red) which is labeled with a calibration bar code.
6. Click “ ” in the Test interface.
QC
1. Take out the QC materials from 2~8℃ and recover the room temperature for about 10~15
minutes.
2. The Lot and target value can be automatically input by scanning the QR code on the external
box of the QC materials with the external barcode reader, or manually input in the QC interface.
3. Add 100uL of hemolytic agent L to dissolve QC materials, shake well and let stand for 30

minutes.
4. Conduct pre-dilution according to the ratio of 10:1500 (add 1.5 mL hemolytic agent L to 10uL
QC materials). Shake well and cut off the tube cap.
5. Put the EP tube with diluted QC materials into the 1st and 2nd position of the calibration rack
(marked orange) which is labeled with a QC bar code.
6. Click “ ” in the Test interface.
Sample Test
■ Routine sample test
1. Load Sample:
1.1 Whole blood mode: Directly put the whole blood sample into the sample rack

(marked white).
1.2 Diluted blood mode: Put the diluted blood sample into the diluted sample holder and
place the sample rack (marked white).
1.3 Anemia mode: Directly put the anemia sample into the Anemia rack (marked blue).
2. Place the prepared rack on the sample loading site.

3. Click “ ” in the Test interface.
■ Emergency sample test
1. Place the whole or diluted blood sample in the STAT port.

2. Click the “Settings” button in the System interface -> Click the “STAT Position” button.
3. Click the “ ” button after return the Test interface when the routine sample
test is running.

4. After the current sample test, the instrument will automatically identify the sample type
and numbers, start the emergency sample test.
Result Remarks
e1:Total area less than 50,000
1. Confirm that the sampling needle is not blocked and the sampling tube is not leaking;
2. If it is a anemia sample, please use the anemia sample rack to rest, or use the dilution
mode to reduce the dilution ratio and retest, such as adjusting from 10:1500 to 20:1500.
e2:Total area more than 270,000
1. Suggested to shake or increase the dilution ratio to retest, such as adjusting from
10:1500 to 5:1500.
e3:Result more than 15%
1. It indicates that the sample may be abnormal, and the operator is recommended to
make a comprehensive judgement based on other clinical indicator.
e4:Result less than 4%
1. It indicates that the sample may be abnormal, and the operator is recommended to
make a comprehensive judgement based on other clinical indicators.

e5:Variants detected
1. When variants are detected, the mutant protein components will be displayed in the "V_
Win" line, and HbA1c results will be marked with " * " and marked red.
e6:ADC415 value reach max value during testing
1. Confirm that the chromatographic column temperature is set correctly (30~40 ℃).
2. In "System" - "Maintenance" interface, select valve A, start the high pressure pump, and
after the system pressure is stable, reconfirm whether the value of ADC415 is between
24000 and 28000.If abnormal, please contact the Lifotronic’s Service Department.
e7:A0 Total area less than 80%
1. It indicates that the sample may be abnormal, and the operator is recommended to
make a comprehensive judgment based on other clinical indicators.
e8:Target peak is out of range
1. Confirm that the chromatographic column temperature is set correctly (35 ℃), and
system pressure is normal(about 5~8MPa).If no,please contact the Lifotronic’s Service
Department.
e9: Pressure is abnormal
1. It indicates that the analyzer may be abnormal, and the operator is recommended to
check the analyzer after finishing test.
e10: TP is too low
1. It indicates that the analyzer may be abnormal, and the operator is recommended to
check the analyzer after finishing test. Then try to run those samples again.
e11: Hb F more than 40% (Thalassemia mode)
1. It indicate that Hb F is more 40%, HBA1c is not reportable by HPLC in H100.
e12: Unknown peak (degradation peak) more than 10%

1. It indicate that unknown peak (degradation peak) more 10%, HBA1c is not report-able
by HPLC in H100.
e13: Suspected HbH
1. It indicate that this sample might be HbH.

Common Troubleshooting Solutions
1. Pressure too low
1.1 Check the remaining volume of reagents.
1.2 Check the connection between the chromatographic column and filter for leakage.
1.3 Check the evacuation valve is tightened.
1.4 If no, loosen the evacuation valve, perform “ bubble elimination” in the Systerm interface,
then tighten the evacuation valve.
2. Pressure too high
2.1 Replace the filter. If the pressure i.e. continue too high, please replace the
chromatographiccolumn.
2.2 If no, please contact the Lifotronic Service Department.
