Joshua A. Fonbuena: Education

Joshua A. Fonbuena is a senior at California State University, Fullerton majoring in Biological Science with a concentration in Molecular Biology and expected to graduate in May 2019 with a GPA of 3.77. He has been conducting research at the Molecular Parasitology Research Laboratory under Dr. Veronica Jimenez since May 2017 analyzing gene expression in Trypanosoma cruzi parasites with and without the TcMscS gene using RNA-seq, qPCR, and bioinformatics. He has presented this research at a campus symposium and received the MARC Scholarship and been on the Dean's List. His skills include PCR, qPCR, RNA isolation, cDNA synthesis, bioinformatics, and

B. S. Biological Science
Concentration: Molecular Biology Cumulative GPA: 3.77

Research Experience
Molecular Parasitology Research Laboratory May 2017 - present
PI: Dr. Veronica Jimenez, Assistant Professor, CSUF
Collecting and analyzing RNA sequencing (RNA-seq) data from Trypanosoma cruzi wild-type
(WT) parasites and knock-out (KO) parasites that have had the TcMscS gene removed, and
selecting gene candidates with the most upregulated and downregulated gene expression to
investigate how parasites compensate for the loss of the TcMscS channel.
 Gene expression analysis of WT and mutant KO parasites by PCR and quantitative PCR
(qPCR)
 qPCR data quantified using ΔΔ quantification cycle (Cq) calculation method, which
calculates relative gene expression levels between the target gene (TcMcS) and
reference gene (GAPDH)
 Extracted RNA from WT and mutant T. cruzi parasites and synthesized cDNA
 Culturing WT T. cruzi parasites and mutant Cas9, knockdown (KD) parasites with
reduced TcMscS genes, and KO mutant constructs created by Dr. Jimenez
 Bioinformatic analyses to determine differential expression of RNA seq data was done
using the Cufflinks suite protocol on LINUX command line
 Currently investigating gene ontology analyses of RNA seq data using RStudio

Hobbies
 Music has always been a big part of my life and has allowed me to connect with other
people. I revel in all aspects of music and take great pleasure in performing with other
musicians. I play the guitar, bass, and a variety of other stringed instruments. I used to
play with a band called The VP and we played shows in the local LA and Orange County
area, including the House of Blues Anaheim.
 I welcome the outdoors and enjoy hiking, exercising, and being active with sports. I have
run a marathon and hope to complete a 100-mile bike race, which is known as a century,
in the near future.
